Monoclonal antibodies against alpha toxin of Clostridium perfringens.

Ten distinct monoclonal antibodies (MAbs) against alpha toxin of Clostridium perfringens were produced by the fusion of SP2/O with spleen cells of mice immunized with alpha toxoid, and alpha toxin mixed with or without ethylenediamine-tetraacetate (EDTA). The antibody activity was evaluated by antigen-binding activity in an enzyme linked immunosorbent assay (ELISA), by phospholipase C (PLC)-neutralizing activity using both egg yolk lecithin and p-nitrophenylphosphoryl-choline (PNPPC) hydrolysis reactions and by anti-lethal activity in mice. Since the toxin-neutralizing activities of each MAb were not parallel, it has been suggested that the three biological activities may not be located in the same site in the toxin molecule. This report also describes the development of a simple purification of the toxin by affinity chromatography and a sensitive immunoassay for quantitation of the toxin using the monoclonal antibody.

Coal mine workers' pneumoconiosis (CWP): in vitro study of the release of organic compounds from coal mine dust in the presence of physiological fluids.

Solvents like dichloromethane generally are used to yield exhausted extraction amounts of the organic compounds in coals. Leaching of coal mine dust by dichloromethane yields extracts with comparable amounts of alkanes, aromatics, and phenolic compounds. Dominantly phenolic compounds are leached from coal mine dust by aqueous solutions saturated in lecithin because of their high water solubility. High concentrations of phenolic compounds can be extracted from coal mine dust generated from low-rank coals. Phenolic compounds leached by fluids adapted to physiological conditions correlate with high cytotoxicities of the dust from low-rank coals. Adaptation of leaching fluids to physiological conditions allows a more realistic estimation of experiments. Coal mine dust with varying coal content of different ranks can be seen as a parameter reinforcing the cytotoxic potential of coal mine dust.

The influence of manufacturing parameters on the formation and growth on autoclaving of a 40% V/V Bis-Perfluorobutylethene emulsion.

The influence of different process variables on the number of large particles before and after autoclaving of a 40% V/V Bis-Perfluorobutylethene emulsion stabilized by egg yolk lecithin, made isotonic with blood, was examined. The concentration of emulsifier, emulsification and autoclaving time and temperature, fill volume and the cooling gradient applied to the emulsion after autoclaving all affect the number of large droplets and hence the stability and acceptability of the finished product. This work suggests that validation of equipment and process to very exacting specifications and strict adherence to specified manufacturing protocol is essential for the reproducible production of fluorocarbon emulsions acceptable for intravenous administration.
